Mit Hilfe der \ac{PCR} konnen Nukleinsäurefragmente gezielt

vervielfaltigt werden. Die Spezifität der Reaktion ist hierbei durch die Auswahl der

Eine PCR gliedert sich grundsätzlich in drei aufeinanderfolgende Phasen: Während

der Denaturierung bei \SI{95}{\celsius} wird der Doppelstrang in die beiden Einzelstränge

aufgespalten. Das Annealing bei \SIrange{50}{60}{\celsius} führt zur Anlagerung der Primer an die

komplementären Sequenzabschnitte der Einzelstränge des Templates. Die exakte

Temperatur ist hierbei von den Schmelztemperaturen der eingesetzten Primer abhängig.

Die \ac{SLIC} \citep{Li2007} macht sich die Exonukleaseaktivität der T4-\acs{DNA}-Polymerase zu Nutze, um 5'-Überhänge zu generieren, welche anschließend in einer enzymunabhängigen Reaktion ligieren.

Der Zielvektor pET-16b wurde zunächst über die Restriktionsenzyme \textit{Nde}I und \textit{BamH}I (New England Biolabs, Frankfurt) linearisiert. Hierfür wurden in einem Gesamtvolumen von \SI{25}{\micro\liter} \SI{4000}{\nano\gram} Plasmid-\acs{DNA} für \SI{2}{\hour} bei \SI{22}{\celsius} nach Herstellerangaben gedaut.

Der linearisierte Vektor wurde anschlieend über ein Agarosegel gereinigt und die \acs{DNA} aus dem Gel gelöst.
|
||||
|
||||
Die für die \ac{PPDK} kodierende Sequenz wurde wie in \ref{sec:pcr} beschrieben amplifiziert und ebenfalls über ein Agarosegel gereinigt.
